Evaluation of mechanical heart function (isolated heart)

Mechanical function of isolated heart

We own an apparatus allowing the evaluation of the cardiac mechanical function in the in vivo and ex vivo situations.

Estimation of myocardial contractility in the in vivo situation is allowed by means of a Millar pressure probe introduced in the left ventricular cavity via the right carotid. It permits the accurate measurement of different parameters characterizing cardiac contraction (left ventricular developed pressure, heart rate, dP/dt max (contractility) and dP/dt min (relaxation)).

Our apparatus also allows the measurement of cardiac function in isolated hearts. The hearts are perfused with an appropriate medium whose temperature and composition are controlled. The mechanical function is determined thanks to a latex ballon introduced in the left ventricular cavity: it is linked to a pressure gauge and allows the determination of the intraventricular pressures during the cardiac cycle. The determined parameters are similar to those evaluated in the in vivo situation. The isolated hearts are also paced electrically at a fixed rate. In these standard conditions, the functional status of the cardiac contractile cells are determined accurately. We can also estimate the functional activity of the coronary vascular cells (endothelial and smooth muscle cells) by determining the coronary micro-vessel reactivity. The isolated perfused heart is interesting because it allows the modelling of several pathologies like myocardial infarction during which the letal arrhythmias can be assessed.
