Multimodal prevention of loss of mobility during inflammatory rheumatisms, identification of biomarkers of muscle health.

Financial support from the Auvergne-Rhône-Alpes region as part of the scientific challenge 3 of I-SITE program (2018-2020): personalized mobility as a key factor of health. Coordinator : Anne Tournadre.

The aim of this project is to use translational approaches ranging from a pre-clinical animal model to a cohort of patients with rheumatoid arthritis to explore the mechanisms of sarcopenia in inflammatory conditions and reduced mobility, to identify and validate therapeutic targets and biomarkers, to develop new innovative multimodal diagnostic, preventive and therapeutic strategies combining targeted therapy and nutrition for the definition of personalized medicine.

In a first phase, in an animal model combining joint inflammation and lipotoxicity induced by a lipid-rich hypercaloric diet, we will 1) explore the mechanisms of muscle lipotoxicity associated with this pathological condition, 2) analyze the impact of specific cytokine inhibitors or JAK/STAT signaling pathways and dietary intake of n-3 polyunsaturated fatty acids (docosahexaenoic acid) in the form of phospholipids extracted from egg products on muscle lipotoxicity, and 3)identify the lipidomic profile  associated with muscle lipotoxicity in animal serum.

In a second phase, this lipotoxic signature will be explored as a biomarker of sarcopenia in a cohort of patients suffering from rheumatoid arthritis. The obtained data will be used to initiate the development of a predictive algorithm and a personalized preventive and therapeutic strategy (4P medicine) for sarcopenia.
